In order to improve the understanding and risk management of clinical transporter-mediated DDIs, the IQ Transporter Working Group completed an extensive data collection project of both in vitro (substrate liability and inhibition) and clinical transporter data (victim and perpetrator DDIs) on IQ member company drugs and new molecular entries. The primary goal of the project was to investigate the in vitro to in vivo correlation of transporter drug interactions for DDI and identify the overall interaction magnitude and clinical implications of the studies. In vitro substrate and inhibition data, including assay conditions, and clinical DDI study data was collected anonymously. The collection included compound characteristics such as physicochemical properties, general available ADME properties including permeability, protein binding, blood to plasma partitioning and PK parameters The demographic data and the in vitro to in vivo correlation of the data will be presented and compared with the current regulatory decision trees.
